Modafinil (Modalert, Modvigil) and Armodafinil (Waklert) are the two dominant wakefulness-promoting agents used off-label for focus, shift-work sleep disorder, and cognitive endurance. Here's how they differ.
Chemistry
Modafinil is a racemic mixture (equal parts R- and S-enantiomers). Armodafinil is the pure R-enantiomer — the longer-acting half. This is the entire basis of the pharmacological differences.
Onset and duration
- Modafinil 200mg (Modalert/Modvigil): onset 30–60 min, peak 2–3 hr, duration 10–12 hr.
- Armodafinil 150mg (Waklert): onset 60–90 min, peak 3–4 hr, duration 12–15 hr.
Subjective feel
- Modafinil: Faster kick-in, more "up" feeling, better morning wakefulness.
- Armodafinil: Smoother, longer, more consistent focus. Preferred for shift-work and extended-duration tasks.
Dose equivalence
Armodafinil 150mg is roughly equivalent to Modafinil 200mg in total wakefulness effect, but with a longer tail.
Side effects (shared)
- Headache (most common — hydrate aggressively).
- Insomnia if taken after noon.
- Reduced appetite.
- Rare: rash / SJS — discontinue immediately if rash appears.
Which to choose
- Morning focus, single peak: Modafinil 200mg.
- Long day, night shifts: Armodafinil 150mg.
- Sensitive to stimulation: Start Modafinil 100mg (half tablet).
Interactions
Both are CYP3A4 inducers — reduce efficacy of hormonal contraceptives (use backup) and some benzodiazepines. Discuss with a prescriber.
